It has been demonstrated (1-3) that shRNA constructs with longer duplexes (27-29mers) provides multiple advantages over the conventional 21mer duplex:
- Up to 100 times more potent than the conventional 21-mer shRNA or siRNA
- Much less shRNA is required for effective knock-down
- Non-detectable interferon induction
- Minimal "off-target" effect
- Accessible for sites that are refractory to 21-mer siRNA.
