Novasep has developed a modified silica-based protein A media for capturing monoclonal and polyclonal antibodies from high titer and large volume fermentation feedstock.
AbSolute is based on a modified silica matrix that exhibits strong mechanical, thermal and chemical stability. The matrix is rigid and incompressible; so a 1 L column is packed with 1 L of AbSolute™ and the media does not shrink or swell in different solutions. These exceptional mechanical properties combined with a unique kinetic performance allow the use of larger columns and greater bed lengths for operation at higher velocities (up to 1000 cm/h). The particles are small (44 µm) with a perfect spherical shape and very narrow size distribution. Pore size is 1000 Å and shows narrow size distribution (+/- 150 Å). Optimized particles and pores allow high efficiency and faster mass transfer which increase the Dynamic Binding Capacity (DBC).
AbSolute remains stable after 150 cycles of repeated alkali washing using 50 mM NaOH with a residence time of 20 minutes.
AbSolute offers a better dynamic binding capacity in comparison with another media. Indeed, breakthrough with AbSoluteTM occurs later than with agarose based media and the breakthrough curve for AbSolute is very steep; this is the result of uniform particles and homogenous pore size.
The optimized particle size and uniform pore size distribution maintain rapid mass transfer, resulting in higher dynamic capacities over a wide range of velocities.
Furthermore, this feature combined with low pressure drop and the rigid nature of the homogenous pore base matrix, delivers high productivity.
