HEA And PPA Hypercel Sorbents
Pall Life SciencesSorbents for Mixed-Mode, Hydrophobic Interaction Chromatography
PPA and HEA Hypercel are members of a family of Pall BioSepra chromatography sorbents designed for direct capture of proteins or contaminant removal from crude feedstreams. These sorbents bind proteins by a mixed-mode mechanism, principally hydrophobic interaction, operating in non-conventional, low-salt conditions (physiological HIC). They are based on the mechanically stable base matrix of Hypercel, currently used in the production of clinical material for late-stage clinical trials (MEP HyperCel).
PPA and HEA HyperCel facilitate binding from feedstocks or intermediate stage product that cannot be directly applied to traditional ion exchange or HIC sorbents:
- Conductivity is too high for traditional ion exchange sorbents
- Would require significant addition of salt for traditional HIC.
- Unlike traditional HIC, the target protein is recovered in dilute buffer, reducing the need for intermediate diafiltration.
Chromatographic behavior is based on a combination of electrostatic and hydrophobic properties of the protein and ligand:
- Can provide unique selectivities not accessible with traditional IEX or HIC.
- The differing retentivities & selectivities of PPA HyperCel, HEA HyperCel and MEP HyperCel can be screened to facilitate process development.
Features and Benefits
