Ajinomoto has developed a new patentable bacteria r-protein expression system which makes large scale production (kilogram) of IGF-1 possible.

The use of IGF-1 in cell culture media can have a major impact on yields when compared to serum insulin or amino acid media. The major drawbacks to IGF-1 are costs, consistency, and impurities. Existing production methods use an E.coli expression system. This production process requires; cultivation, cell harvest, and a complex purification process. The E.coli process is hindered by cell disruption and refolding. Cell disruption requires purification which is a complex process. Refolding is an issue with E.coli and can reduce active IGF-1 yield to 50%. The result is an expensive product, with inconsistent yields, and the potential for impurities.

Ajinomoto’s new innovative process uses a unique expression system, corynebacterium glutamicum (amino acid producer). With this breakthrough system the production process consists of cultivation, cell removable, and a simple purification process. Refolding and cell disruption are very limited which makes the purification process very simple. The system produces purified active IGF-1 in relatively large scale production. The system is cost efficient and produces a IGF-1 that is 100x more active than serum insulin. This new expression system makes IGF-1 a viable option for cell culture operations not currently using IGF-1 in their cell culture operation. It also has the potential to reduce costs for operations currently using other IGF-1’s.
