The growth environment of the roller bottle is very dynamic when compared to static culture systems. Cells are intermittently bathed in the nutrient media through the continuous, slow rotation of the bottle. For the majority of the cycle that the cells are not submersed, a thin layer of media coats the cells providing an increase in the media surface coverage relative to its volume. This increase in overall growth area, combined with the higher media surface to volume ratio and the gentle agitation, boosts gas exchange and enhances monolayer formation.
