Process Optimization
Source: XcellerexBioreactor
Xcellerex's high-throughput (HT) bioreactor optimization platform may be used to screen various bioreactor parameters such as temperature, pH, dissolved oxygen and feed strategy leading to significant improvements in cell culture yields and product quality. Xcellerex can optimize these parameters, including investigation of pH and temperature shifts, in order to improve yields or solve specific process problems.
In combination with bench scale bioreactors, this technology will allow the rapid evaluation of bioreactor parameters which may impact yield and product quality.
HT Bioreactor Optimization
- Optimize temperature, pH, DO set points
- Assess impact of temperature and pH shifts
- Confirm scalability of cell line, media, feed and process parameters
Purification
Xcellerex's high-throughput (HT) purification screening platform may be used to screen various resins, binding, wash and elution conditions as well as the sequence of binding chemistries, and can investigate thousands of experimental conditions in parallel, allowing the range and interaction of process variables to be thoroughly understood. Each experimental condition may be evaluated based on yield using the HT protein assay using other HT assays. The most promising purification conditions can be confirmed on traditional lab scale equipment.
Xcellerex developed this approach based on the same platform of automation and procedures used for HT media optimization. Purification processes are traditionally developed on lab scale equipment based on conditions developed for previous processes in order to minimize development time.
HT Purification Screening
- Rapidly screen different resins
- Optimize binding, wash and elution conditions
- Optimize the sequence of binding chemistries
- Analyze resin lifetime, leaching, cleaning efficiency
Formulation
Xcellerex's high-throughput (HT) excipient screening platform evaluates promising product formulations using the same robotic platform and procedures used for HT media optimization. Different excipients can be tested at varying concentrations, pHs, ionic strengths and different product concentrations in thousands of parallel conditions. These formulation conditions may be subjected to stresses (e.g. high temperature, high oxygen tension, agitation) to accelerate product destabilization, and assessed for product stability.
HT Excipient Screening
- Screen a broad range of excipients
- Optimize excipient and product concentrations
- Rapidly evaluate pH, ionic strength
